A pilot study on the association of KCNJ11 genetic polymorphism (rs5215 Val250Ile) with Type 2 diabetes mellitus among the Saudi Arabian population (Eastern province)

Abstract
Background: Diabetes is a chronic metabolic disorder. Single nucleotide polymorphisms in the KCNJ11 gene have shown to play a role in the development of Type 2 diabetes mellitus (T2DM). The present pilot study was aimed at evaluating the relationship between KCNJ11 polymorphism (rs5215) with T2DM susceptibility in the eastern province among the Saudi Arabian population.
Methodology: A case-control study was conducted on 49 confirmed T2DM patients and 39 control subjects from the family and community center in Dammam. The T2DM diagnosis was based on the American Diabetic Association criteria. The peripheral blood was collected by venipuncture. The genomic deoxyribonucleic acid was extracted and the KCNJ11 gene rs5215 polymorphism was genotyped by TaqMan chemistry using real-time polymerase chain reaction. Statistical tests were performed using statistical package for the social sciences ver19 software and the results were counted as significant if the p-value is <0.05.
Results: Anthropological parameters, such as weight and body mass index, in the patients and control groups were significantly different (p = 0.012 and p = 0.006); respectively. The minor allele “C” of rs5215 was overrepresented in T2DM patient (0.24) compared to controls (0.15), the association did not reach statistical significance. The rs5215 genotype (p = 0.443) and allele (p = 0.139) showed an insignificant relationship with the T2DM.
Conclusion: This study is the first of its kind to report the frequency and to assess the relationship between rs5215 of KCNJ11 with T2DM in the eastern province of the Saudi Arabian population. The rs5215 of the KCNJ11 gene showed no significant association with the T2DM in this population.
